Resveratrol attenuated high intensity exercise training-induced inflammation and ferroptosis via Nrf2/FTH1/GPX4 pathway in intestine of mice

Background/aim: Moderate exercise has beneficial effects for human health and is helpful the protection against several diseases. However, high intensity training caused gastrointestinal syndrome. Resveratrol, a plant extract, plays vital role in protecting various organs. whether resveratrol protected mice training-induced intestinal damage remains unclear. In this study, our objective was to investigate protective mechanism of training-treated mice. Materials methods: Mice were treated with swimming protocol and/or (15 mg/kg/day) 28 consecutive days. Then, sacrificed, series evaluation indicators, including inflammatory factors permeability gut, measured based on model. The expressions (tumor necrosis factor (TNF)-α; interferon (IFN)-α, interleukin (IL)-6 IL-10), oxidative stress (Nrf2, glutathione (GSH), hydrogen peroxide (H2O2), catalase (CAT) malondialdehyde (MDA)), barrier (gut permeability, ZO-1, Occludin Claudin-1 as well ferroptosis (Fe2+, Fe3+, SLC7A11, peroxidase 4 (GPX4) ferritin heavy chain 1 (FTH1)) measured, respectively. Results: High induced colon damage, manifested inflammation (increased TNF-α, IFN-γ IL-6 concentrations, decreased IL-10 concentration), (the increase H2O2 MDA concentration, reduced CAT GSH activities), injury gut fatty-acid binding protein inhibited expressions) increased Fe2+ Fe3+ suppressed phosphorylated Nrf2, GPX4 FTH1), which relieved by treatment Conclusion: Resveratrol attenuated through activating Nrf2/ FTH1/GPX4 pathway mouse colon, providing new ideas prevention occupational disease athlete.
